Following contraction, the release of calcium from intracellular storage sites aids in the relaxation of skeletal muscles. In order for the actin and myosin filaments to interact and produce contraction, calcium ions must first bind to troponin after being released from the sarcoplasmic reticulum during muscle contraction. Calcium must be actively moved back into the sarcoplasmic reticulum, lowering its concentration in the cytoplasm, for relaxation to take place. Because troponin releases calcium as a result, tropomyosin can cover the actin binding sites, halting the contraction and causing the muscle to relax.
